The Suppressive Effect of 6-Mercaptopurine on the Antibody Production by the Time of Administration

Abstract
Random bred albino rabbits of either sex weighing 2 to 2.5.kg were injected intravenously with 50 mg egg albumin in 5 ml saline two times for two days. 6-mercaptopurine administration was started at 48¡©hour before or after the first injection of the antigen and simultaneously with antigen and these procedure were continued for five successive days. Partial bleeding of pre-immunization and successive bleedings (on day 6, 11, 16, 21, 26, 31 and 36) after the first immunization were made from the marginal ear vein. Precipitation, passive hemagglutination, modified hemolytic reaction, and complement fixation reactions were applied for the titration of antibodies in sera, The suppressive effect of 6-mer¡©captopurine on the antibody response of immunized rabbits were studied for the anti-egg-albumin rabbit sera. Results were summarized as follows:
1. Antibody production was markedly suppressed when administration of 6-mercaptopurine was started at 48-hour before or simultaneously the first injection of antigen.
2. Antibody production was less marked when administration of 6-mercaptopurine was started at 48¡© hour after the first injection of antigen.
3. The suppressive effect of 6-mercaptopurine on the production of precipitin and complement fixing antibodies were rather significant than passive hemagglutinin and modified hemolysin titers.
